Carnation Leaf Agar (CLA)
Add several sterile dried carnation leaf pieces, preferably gamma-irradiated, to Petri dishes just prior to addition of 10 to 15 mL of TWA.
Czapek Yeast Extract Agar (CYA)

Method
- Add 1.0 mL/L of trace metals solution.
- Sterilise by autoclaving at 121°C for 15 minutes.
(Trace metals solution: 1% w/v ZnSO4.7H2O and 0.5% w/v CuSO4.5H2O in distilled water).

Method
- Add minor constituents and agar to 500.0 g distilled water and steam to dissolve agar.
- Add glucose rapidly while stirring.
- Sterilise by steaming for 30 minutes. Do not autoclave.
Malt Extract Yeast Extract 5% Salt 12% Glucose Agar (MY5-12)

Potato Dextrose Agar (PDA)
Ingredients

Potatoes | 250.0 g |
Glucose | 20.0 g |
Agar | 15.0 g |
Water, distilled | 1.0 L |
Method
- Dice unpeeled potatoes into 500 mL of water
- Steam or boil for 30 to 45 minutes.
- Melt agar in 500 mL of water.
- Strain potato through several layers of cheese cloth into a flask containing the melted agar.
- Squeeze some potato pulp through the cheese cloth as well.
- Add the glucose and mix thoroughly, make up to 1 L with distilled water.
- Sterilise by autoclaving at 121°C for 15 minutes.
